摘 要:本文初步建立了HPLC(High performance liquid chromatographic method高效液相色谱法)测定高油辣椒制品中辣椒制品中两种主要辣椒素类物质的方法。样品以甲醇+正己烷进行预处理,采用反相高效液相色谱分离,紫外检测器;根据色谱峰的保留时间定性,以外标峰面积法定量辣椒素(C)、二氢辣椒素(DC)。采用色谱柱,ZORBAX SB-C18;流动相,V(甲醇)∶V(水)=65:35;UV检测波长,280 nm;辣椒素和二氢辣椒素RSD分别为5.11%,5.08%(n=5);回收率分别为89.4%、90.0%。辣椒素和二氢辣椒素在20-200ug/ml范围内线性良好。方法满足辣味水平评价要求。

DETERMINATION OF CAPSAICIN IN THE HIGH OIL PEPPER PRODUCTS
Abstract:A method was preliminary developed for the determination of capsaicinoids in highly oil-containing pepper productsby high performance liquid chromatography. The sample was extractedwith Methanol and Hexane. The analysis wasperformed on a C18 column (250×4.6 mm i.d.5μm) with 65∶35 Methanol-water as the mobile phase at a flow rate of 1.2 mL/min and detected at 280 nm. The determination of capsaicin(C),dihydrocapsaicin(DC) were processed by peak area andexternal standard method. The precisions of C and DC were good with the RSD of 5.11% and 5.08% espectively (n=5).The recoveries for C and DC are 89.4% and 90%respectively. The two ompounds demonstrated good linear relationship in the range of 20-200 ug/ml.
